Whole body plethysmography 全身體積描記器 - *肺功能檢測(cè)系統(tǒng)
全身體積描記系統(tǒng)(Whole Body Plethysmograph System)用于動(dòng)物在清醒、無(wú)束縛狀態(tài)下測(cè)量動(dòng)物的各項(xiàng)肺功能參數(shù)。該系統(tǒng)簡(jiǎn)單、易用、快速、數(shù)據(jù)輸出量大的特點(diǎn)使其成為國(guó)外藥企研發(fā)機(jī)構(gòu)喜愛(ài)的研究工具主要用于哮喘等新藥研發(fā)過(guò)程中進(jìn)行粗篩實(shí)驗(yàn)。其指標(biāo)參數(shù)能夠良好地反映動(dòng)物對(duì)藥物產(chǎn)生的氣道高反應(yīng)情況適合于需要?jiǎng)游镌谧匀坏臓顟B(tài)下呼吸的研究以及*跟蹤實(shí)驗(yàn)。
每個(gè)體描箱都具備霧化攻擊、溫濕度補(bǔ)償接口另外還具有供水接口和氣體采樣接口以適用于各種特殊需要的研究。
WBP全身體積描記器的主要工作原理
WBP主要檢測(cè)由動(dòng)物鼻部氣流和胸腹呼吸運(yùn)動(dòng)引起的氣流變化導(dǎo)致的"box flow"數(shù)據(jù)變化并參考溫度、濕度和壓力的數(shù)據(jù)計(jì)算得出各種指標(biāo)參數(shù)。
將乙酰甲*(methacholine,Mch)或生理鹽水(NS)等通過(guò)霧化器霧化入體描箱內(nèi)箱內(nèi)動(dòng)物吸入霧化氣體后呼吸運(yùn)動(dòng)的改變引起箱內(nèi)壓力、流量產(chǎn)出相應(yīng)的變化連接于體描箱的壓力傳感器(流量傳感器)將采集到的信號(hào)傳至信號(hào)處理系統(tǒng)并在信號(hào)放大器作用下轉(zhuǎn)化為呼吸動(dòng)力學(xué)各指標(biāo)值。
全身體積描記系統(tǒng)可同時(shí)連接6個(gè)體描箱相應(yīng)的傳感器可以將各個(gè)箱內(nèi)壓力、流量的變化同步傳至信號(hào)處理系統(tǒng)處理因此可以同步為多只實(shí)驗(yàn)動(dòng)物進(jìn)行肺功能檢測(cè)。

Whole body plethysmography is an application whereby ventilatory parameters are measured while the subject is conscious and unrestrained. By the use of bias airflow supplies, CO2 levels, temperature and humidity are maintained at normal levels enabling measurements to be made over several hours.
During inspiration there is a slight net pressure rise within theplethysmograph due to increased temperature and humidity of the air as it enters the subject’s lungs (gas conditioning). During normal breathing, this pressure difference is directly proportional to the respiratory pattern of the subject, allowing the researcher to measure respiratory rate and tidal volume, in accordance with ICH S7 Guidelines.
The system is commonly used in airway hyper-reactivity studieswhere, in addition to changes in ventilatory parameters, valuable data can obtained
by quantifying changes in the shape of the respiratory waveform, e.g., Penh. However, it should be noted that when bronchoconstriction is present,
the respiratory signal is dominated by the effects of gas compression and therefore exact measures of tidal volume can not be determined.
